NEOGEN Iron Sulphite Agar, 500 g
Iron Sulphite Agar is a medium for the detection of thermophilic anaerobic organisms causing sulphide spoilage in food, and is not intended for use in the diagnosis of disease or other conditions in humans.This formulation is a modification of Cameron Sulphite Agar, which was developed by the National Canners Association of America (now the Grocery Manufacturers Association). Iron Sulphite Agar has a reduced concentration of sodium sulphite to allow improved detection of some strains of Clostridium sporogenes. Beerens, and later Mossel, demonstrated that some strains of C. sporogenes would not tolerate sodium sulphite levels of 0.1%. This organism has the ability to cause human illness and death, particularly in immunocompromised individuals and pregnant women. Epidemiological evidence from outbreaks of listeriosis has indicated that the principle route of transmission is via consumption of foodstuffs contaminated with Listeria monocytogenes. Implicated vehicles of transmission include turkey frankfurters, coleslaw, pasteurized milk, Mexican style cheese, and pate. Dextrose Tryptone Broth is used for cultivation of mesophilic or thermophilic spoilage microorganisms from food in a laboratory setting. Dextrose Tryptone Broth is not intended for use in the diagnosis of disease or other conditions in humans.In the 1930’s, the National Canners Association specified use of Dextrose Tryptone Agar for isolating “flat sour” organisms from food products. “Flat sour” spoilage of canned foods is caused by Bacillus coagulans (Bacillus thermoacidurans). Bacterial growth results in a 0.3 – 0.5 drop in pH, while ends of the can remain flat. B. coagulans is a soil microorganism and found in canned tomato products and dairy products. Deibel, Evans, and Niven tested thiamine-requiring bacteria, specifically Lactobacillus viridescens. Their formulations led to the development of APT Agar.Lactic acid bacteria, a group of acid-producing bacteria, include the genera Streptococcus, Leuconostoc, Pediococcus, and Lactobacillus. These organisms are widespread in nature, associated with bacterial spoilage of foods including dairy products, meat, and vegetables. Rappaport-Vassiliadis Medium with Soya is used as a selective enrichment medium for the detection of Salmonella spp. in food, animal feed and in environmental samples from the food production area as described in ISO 6579-1:2017. RVS is not intended for use in the diagnosis of disease or other conditions in humans.Rappaport et al. originally described the broth, which was later modified by Vassiliadis et al. and optimized by van Schothorst et al. Enzymatic digest of soya provides a source of nitrogen and amino acids and sodium chloride provides osmotic pressure. The potassium phosphate components act as a buffer. Magnesium chloride and malachite green act as selective agents against non-target microorganisms. Blood Agar Base No. 2 is used with blood for the isolation and cultivation of a wide variety of fastidious microorganisms. The medium gives colonial appearances, hemolysis patterns and pigment production of indicative value.
